Serial monitoring of immunologic function and phenotype of lymphocytes in the blood of transplanted patients randomized to cyclosporine or FK 506.

چکیده
EFFECTS of the drugs FK 506 and CyA on lymphocyte function have been mostly studied in vitro and in vivo in animal models. 1-5 Little information is available about changes in the immune system of patients who, having undergone orthotopic liver transplantation (OL T), are treated with immunosuppressive drugs. To evaluate cellular immunity of patients treated for prolonged periods with FK 506 or CyA, serial immunologic monitoring was performed in liver transplant recipients. The purpose of this monitoring was to compare long-term effects of Cy A and FK 506 on immunologic function and absolute numbers of circulating lymphocyte subsets in liver transplant recipients, to determine if these drugs had similar or different immunosuppressive effects in vivo.

Immunosuppression switch in pediatric heart transplant recipients: cyclosporine to FK 506.
OBJECTIVES We studied rejection, allograft function and side effects, such as hypertension, renal dysfunction and hypercholesterolemia, in seven patients switched from cyclosporine-based triple-drug immunosuppression to FK 506. BACKGROUND A subset of pediatric heart transplant recipients treated with triple-drug immunosuppression consisting of cyclosporine, azathioprine and prednisone experie...
متن کاملRandomized trial in primary liver transplantation under immunosuppression with FK 506 or cyclosporine.
THE USE of cyclosporine (CyA) has greatly impacted the success of liver transplantation. FK 506 is a newly described immunosuppressive agent, with mechanisms of action similar to those of Cy A, but more potent on a weight-ta-weight basis. Pilot studies of FK 506 in primary liver transplantation have shown encouraging results. 1 A randomized trial using FK 506 with low-dose steroids (LDS) was co...
متن کاملCorrelation between bioassayed plasma levels of FK 506 and lymphocyte growth from liver transplant biopsies with histological evidence of rejection.
FK 506, a novel immunosuppressive agent, was first used to reverse rejection in liver recipients who had failed to respond to conventional therapy.' The successful outcome of the initial study has led to clinical trials of FK 506 as the primary immunosuppressive agent for liver allografts and other organ transplants. 2-4 Ongoing studies include dosage protocols for optimal immunosuppression wit...
متن کاملThe use of FK-506 for small intestine allotransplantation. Inhibition of acute rejection and prevention of fatal graft-versus-host disease.
Small intestine allotransplantation in humans is not yet feasible due to the failure of the current methods of immunosuppression. FK-506, a powerful new immunosuppressive agent that is synergistic with cyclosporine, allows long-term survival of recipients of cardiac, renal, and hepatic allografts. This study compares the effects of FK-506 and cyclosporine on host survival, graft rejection, and ...

Immunosuppressive drugs are now unavoidibly used after the transpantation of liver, heart and bone marrow, as well as in certain autoimmune diseases.among these agents,cyclosporine-A (CYA) is one of the most effective Immunosuppressants which is widely used.but as any Immunosuppressive agent, the use of CYA is not without harmful side effects.neurotoxin, nephrotoxic and hepatotoxic complications,m...
